Chromatographic area and analyte content: two different questions

Chromatographic area, chemical identity and analyte content answer different questions. A useful report states which property was measured and how the method supports it.

What a peak-area percentage describes

An HPLC/DAD chromatogram records the detector response of components that are separated and detected under specified conditions. A relative peak-area percentage describes the integrated response assigned to a peak compared with the included integrated response. The method, wavelength and integration choices matter.

Different components may have different responses, may co-elute or may not be detected under that method. A high principal-peak area percentage therefore does not automatically establish total mass purity, complete molecular identity or milligrams of analyte per vial.

Why weighing powder answers a different question

A balance measures the gross mass placed on it. A research powder may contain water, counterions, residual solvents or other constituents. Gross powder mass does not, by itself, establish the mass of the named analyte.

What a quantitative-content claim needs

  • An exact analyte and a defined reporting basis.
  • An appropriate qualified reference material and a defensible assigned value.
  • Suitable preparation, recovery, calibration and controls.
  • Demonstrated method performance for the material and the claim.
  • A calculation record, units and relevant limitations.

Read a result with its method

Check the sample identifier, the measured property, the units, the method/version and the basis of comparison. Keep chromatographic profile, identity evidence, gross powder mass and analyte content separate unless each claim has supporting evidence.
